Cốt truyện chính của tựa game nói về một nhóm khủng bố dẫn đầu bởi Sergei Dekker. Vào đầu game nhóm khủng này đã ăn cắp được 4 vũ khí hạt nhân tại một cơ sở lưu trữ tại Nga và đem chúng đi rêu bán cho các nước khác nhau đổi lại với số tiền đó chúng đi mua hàng loạt các loại vũ khí hủy diệt hàng loạt. Và người chơi chúng ta vào vai John Mullins, làm việc tại tổ chức lính đánh thuê tại Mỹ (Soldier of Fortune), cùng với người đồng đội của anh là Aaron "Hawk" Parsons ngăn chặn âm mưu của lũ khủng bố và tránh việc các vũ khí hạt nhân kia rơi vào nhầm ta kẻ xấu. 

Xuyên suốt game người chơi sẽ được đi đến các địa điểm như New York, Sudan, Siberia, Tokyo, Kosovo, Iraq, Uganda và cuối cùng là đại bản doanh của nhóm khủng bố đóng chốt tại Đức.

Soldier of Fortune nổi tiếng về mặt bạo lực của game, khi khả năng phá hoại của các vũ khí trong game đều có thể làm lộ ra "lục phủ ngũ tạng" của cơ thể con người và tương ứng với các bộ phận khác nhau có các kiểu phá hoại khác nhau . Chính đặc điểm này là điểm thu hút chính của tựa game, giống như khả năng phá hoại môi trường ở Red Fraction hay là bullet-time của dòng game Max Payne vậy.
